Abstract
The treatment options for migraine are limited and many patients experience a lack of efficiency or intolerable side-effects from the current treatment. Hence, the development of new antimigraine drugs is essential. The development and testing of new migraine drugs is, however, cumbersome and time-consuming. Therefore, the overall aim of the present thesis was to develop a pragmatic human model which can be used to test new drug candidates for migraine. We conducted a series of double-blinded, randomized, placebo-controlled, cross-over studies using 200mg cilostazol (a phosphodiesterase-3-inhibitor) to induce headache/migraine. The headache/migraine were treated with sumatriptan (a specific acute anti-migraine drug) in different formulations. Thirty participants (either healthy or patients with migraine without aura (MO)) were enrolled in each study. The patients filled out a self-reported headache questionnaire until 12 hours after cilostazol. Cilostazol induced headache with migraine-like features in most healthy volunteers and in all MO patients. Sumatriptan tablets showed only a trend towards efficacy in healthy volunteers (study 0, my master assignment). Sumatriptan tablet pre-treatment (study I) prevented the headache from developing in healthy volunteers as intended but the placebo group did not develop much headache and thus the study was statistically negative....
