Reverse phase HPLC – Analysis and separation of complex chemical mixtures
High-performance liquid chromatography (HPLC), is used to identify, quantify and separate components in a chemical mixture. Pumps pass a pressurized liquid solvent containing the sample mixture through a column filled with a solid adsorbent material. Each component in the sample interacts slightly differently with the adsorbent material, causing different flow rates for the different components and leading to the separation of the components as they flow out of the column. HPLC can be used for analysis or separation of the components. HPLC is distinguished from traditional liquid chromatography because operational pressures are significantly higher (50–350 bar) and the resolving power significantly better. The instrument is an Agilent 1100 equipped with a UV/Vis, photodiode array (PDA), a binary pump and a column oven.

Ion Chromatography HPLC – Separation of molecules based on ion interaction
Ion chromatography is a special form of HPLC that allows the separation of ions and polar molecules based on their affinity to an ion exchange column. It works on almost any kind of charged molecule - including proteins, nucleotides, carbohydrates and amino acids. It is currently configured for analysis of carbohydrates at high pH values, which is necessary for ionizing the molecules, but is flexible and can be configured for other purposes. The instrument is a Dionex IC6000 with thermostated autosampler, quarternary pump with degasser, CarboPack column and an electrochemical detector. It operates up to 5000 psi.

FPLC – Fast protein isolation
Fast protein liquid chromatography is used for analysis and separation of protein mixtures, often generated in a protein expression system. The instrument is a highly flexible ÄKTAFPLC (GE Healthcare, Pharmacia) purification system where any custom method (ion exchange, metal-affinity etc.) can be designed through UNICORN software (Cytiva). The system is connected to a PC from where all operation is handled. It has three detectors: an UV absorbance detector a pH detector and a conductivity detector. In contrast to HPLC, the buffer pressure used is relatively low, typically less than 5 bar, but the flow rate is relatively high, typically 1-5 ml/min (up to 20 ml/min). FPLC can be readily scaled from analysis of milligrams of mixtures in columns with a total volume of 5 ml or less to industrial production of kilograms of purified protein in columns with volumes of a liter.
